Dendritic cells (DCs), named for their projections in the form of a "tree" or dendrites, they are responsible for the initiation of responses adaptive immune systems, and function as “sentinels” of the immune system. Paul Langerhans, in 1868, was the first to describe DCs on human skin, although he believed that they were skin nerve cells. DCs are marrow derived leukocytes bone and the most potent type of antigen-presenting cell. They can expand in vitro from bone marrow and blood samples using combinations of various factors growth, such as colony stimulating factor. The manipulation of dendritic cells allows them to become the most effective adjuvant to increase the immune defenses of the host. In the case of cancer, tumors have been shown to suppress DCs by releasing of anti-inflammatory cytokines such as IL-10, thus conditioning the dendritic cells of the medium to originate suppressor T cells. To reverse these mechanisms, you can generate Ex vivo DCs challenged with tumor antigens and reinjected to boost immunity of the host versus tumor cells. Vaccines for DCs generated by this procedure are safe with few side effects, and have been shown to be a possible and effective process in some patients.
